How do I know when to use water vs. PBS as a solvent for diluting a concentrated reagent?

I want to dilute certain stock reagents but I do not know when to use water or phosphate buffered saline (1X PBS) as my solvent. for example:

formaldehye
bovine serum albumin
Triton

For each of these, use water or PBS? is there a general rule I can follow to decide which solvent to use?

Depends on the pH of system that certain compounds are denatured, like proteins. other nucleophiles compete with water when you have to make an organic reaction or maybe its solubility is poor. for those cases, you should use the buffer. it helps to maintain an equilibrated solution.

Well, yeah, that or something along those lines would seem to have been more fair, but the way it was done was the way it was done.  The National League owners were a notably arrogant, hard-headed bunch.

For what it’s worth, neither the Mets nor the Colt .45s were at all happy with the terms of the draft, not so much the structure of it but simply the quality of the players made available.  Indeed, Paul Richards was far more upset about it; in typical bombastic Richards fashion, prior to the draft he proposed to Weiss that both expansion teams boycott the draft, and just stock their rosters via minor league free agents, the waiver wire, and such.

Weiss wouldn’t agree to go along with that, and the National League issued an edict to Richards to quit bitching publicly.  Richards complied up until the draft, but as soon as it was completed, he marched into the press room and announced, “Gentlemen, we’ve just been f—-ed.”
